Also, I wasn't quite clear on one thing: as far as I know, there is currently no way to re-form DNA from restriction digested fragments. Restriction enzymes work by cutting the DNA strand at any location where a given short sequence occurs (for example, one enzyme may cut through wherever this sequence appears:

GTTCCT
CAAGGA

That means that all instances of the above sequence in a genome (and given the sheer number of nucleotides we're talking about, this will happen quite often, sometimes multiple times within individual genes) will be cut, and there is no way to get them to reattach exactly as they were originally arranged. Any of the fragments will bind to any other fragment just as readily (more or less), if DNA-synthesizing enzymes are added to the mixture.
